Good how to question

I have a Mytee M5 portable and have run 75ft of house and still have good suction, if i were to run more than that i think i would use my vacuum booster-I'd like to leave machine outside of house and can use the auto pump out no problem but as far as filling the fresh water tank and using the auto fill i'd like to have like a 40gal fresh water tank inside my van, so how, if possible do i set that up so the water from 40gal tank feeds into fresh water tank on my porty

Quick disconnect to machine, add a valve with float ball shut off to the interior. Use Ross's pump to push water in until the water level shuts off the float.

As much jiggling as a portable gets you'll want to check the float assembly before each use. The good thing is it's outside so if it does malfunction will only spill onto the ground and not onto flooring.

I do agree with Brian, it might be more effort than it's worth.

Although you could hook it up to run from their faucet but you'll probably want to use a regulator to reduce some of the pressure.
