Should I remove rubber mat from cargo area of Chevy Express before installing turckmount equipment ?
I wouldn't. The mat will probably trap water causing rust. Anywhere you drill holes should be cleaned and protected with some type of sealer or rino liner to prevent rust.
Just my opinion.
I don't have any mats in my cargo area.

3/4” rubber flooring has worked best for me. I ordered the mat that Chip Garber recommended, and while it fits nice and is easy to install, I found that it tears easily and some slats are needed to lay in the vans ribbed floor , to facilitate easy movement of wheeled machinery such as my 175, pile lifter etc.

I prefer the Line-X professionally applied product. Under the machine, I use heavy duty rubber matting to absorb the vibration. The rest of the van is just Line-X. Makes it easy to park on a slope and rinse out the van.
